BioLineRx and Hemispherian show preclinical synergy of GLIX1 with PARP inhibitors in HR‑proficient ovarian cancer

BioLineRx Ltd. and its partner Hemispherian AS presented preclinical data at the 2026 ESMO Congress showing that GLIX1, a TET2 activator, synergises with olaparib, veliparib and niraparib in homologous‑recombination‑proficient ovarian cancer models. In a 28‑day patient‑derived xenograft study, low‑dose GLIX1 (50 mg/kg) combined with olaparib (10 mg/kg) produced statistically significant tumour‑growth inhibition, matching cisplatin and outperforming each agent alone at their optimal doses. The companies plan to add an ovarian‑cancer arm to the ongoing Phase 1/2a GLIX1 trial in glioblastoma.

The companies say the data suggest GLIX1 could broaden the use of PARP inhibitors to HR‑proficient ovarian tumours, a large unmet‑patient segment, potentially increasing future market opportunity for GLIX1. If the planned ovarian‑cancer arm proceeds, it could drive additional clinical‑development costs and affect BioLineRx’s valuation.

  • 1GLIX1 combined with three PARP inhibitors showed ZIP synergy scores ranging from 10.9 to 43.2.
  • 2In a 28‑day HR‑proficient ovarian cancer PDX model, GLIX1 50 mg/kg daily plus olaparib 10 mg/kg daily gave statistically significant tumour‑growth inhibition versus vehicle control.
  • 3The low‑dose combination achieved tumour‑growth inhibition comparable to cisplatin and greater than GLIX1 250 mg/kg daily or olaparib 50 mg/kg daily given alone.
  • 4GLIX1 is currently in a Phase 1/2a trial for glioblastoma with three dose‑level patient cohorts enrolled.
  • 5BioLineRx plans to add an ovarian‑cancer arm to the expansion phase of the ongoing GLIX1 trial (NCT07464925).
