Pfizer reports robust Phase 2 results for its 25‑valent pediatric pneumococcal conjugate vaccine

Pfizer announced detailed Phase 2 data for its investigational 25‑valent pneumococcal conjugate vaccine (PF‑07872412, 25vPnC) in healthy infants. The four‑dose regimen generated strong immune responses against all 25 serotypes, including five new serotypes, and showed a safety profile comparable to the approved PREVNAR 20 vaccine. The candidate produced an 8.8‑fold higher GMC for serotype 3 after Dose 3 and about a 15‑fold increase after Dose 4, and functional OPA responses were observed for all serotypes. Pfizer said the data support ongoing Phase 3 trials that aim to protect against roughly 84 % of invasive pneumococcal disease cases in U.S. children under five.

The company expects the Phase 3 program to confirm the broad protection, which could expand the market share of its pediatric pneumococcal portfolio if successful. Investors should watch for Phase 3 enrollment progress and any regulatory feedback that could affect future launch timing.

  • 1The Phase 2 study evaluated PF‑07872412 (25vPnC) in infants using a four‑dose series.
  • 2Robust immune responses were observed against all 25 serotypes one month after Dose 3 and Dose 4.
  • 3The vaccine elicited an 8.8‑fold higher GMC for serotype 3 after Dose 3 and approximately 15‑fold higher after Dose 4 compared with PREVNAR 20.
  • 4Serotype 3 accounts for 10.6 % of invasive pneumococcal disease in U.S. children under five.
  • 5The five new serotypes (15A, 23A, 23B, 24F, 35B) are estimated to add about 30 % of pediatric invasive disease cases compared with PREVNAR 20.
  • 6Phase 3 studies began earlier in 2026 with up to 6,830 participants randomized to 25vPnC or PREVNAR 20.
