Amgen’s IMDELLTRA plus AstraZeneca’s IMFINZI cuts death risk by 58% in extensive‑stage small cell lung cancer

Amgen reported that its DLL3‑targeted bispecific antibody IMDELLTRA, when combined with AstraZeneca’s PD‑L1 inhibitor IMFINZI, reduced the risk of death by 58% (hazard ratio 0.42) in a Phase 3 first‑line maintenance trial for extensive‑stage small cell lung cancer. Median overall survival improved to 21.1 months versus 12.7 months with IMFINZI alone, and median progression‑free survival was 7.4 months versus 1.9 months. The study enrolled 563 patients and showed a safety profile consistent with the individual drugs.

Amgen expects the data to expand IMDELLTRA’s market opportunity, noting Q2 sales of $288 million, up 115% year‑over‑year, and the potential to become a blockbuster therapy. AstraZeneca benefits from partial funding and the prospect of increased durvalumab use in combination.

  • 1The trial showed a 58% reduction in death risk (hazard ratio 0.42; 95% CI 0.32‑0.54).
  • 2Median overall survival was 21.1 months with the combo versus 12.7 months with IMFINZI alone.
  • 3One‑year survival was nearly 80% with the combo compared with 53% for IMFINZI alone.
  • 4Median progression‑free survival was 7.4 months versus 1.9 months.
  • 5563 patients were randomized 1:1 to the combination or IMFINZI alone.
  • 6Grade 3 or higher treatment‑related adverse events occurred in 20.9% of combo patients versus 10.8% of IMFINZI‑only patients.
