Pfizer reports positive Phase 3 progression-free survival result for ELREXFIO

Pfizer said its Phase 3 MagnetisMM-32 trial of ELREXFIO met its progression-free survival endpoint in relapsed or refractory multiple myeloma after anti-CD38 therapy and lenalidomide. At the interim review, over three-quarters of treated patients were alive without worsening disease, with estimated 18-month PFS of 68%; median PFS was not yet reached. The 523-patient study compared ELREXFIO with three established regimens, and Pfizer plans submissions to health authorities.

The result may support Pfizer’s plans to seek broader regulatory use of ELREXFIO in this treatment setting. Pfizer plans to submit the findings globally, while overall survival data remain immature and detailed results are pending.

  • 1MagnetisMM-32 enrolled 523 adults across 26 countries.
  • 2An estimated 68% of patients remained progression-free at 18 months, while median PFS had not yet been reached.
  • 3Grade 3 or higher infections occurred in 27.5% of ELREXFIO patients versus 18.1% in the investigator-choice group.
  • 4Pfizer plans to submit the trial data to global health authorities and present detailed findings at a future medical meeting.
  • 5The trial compared ELREXFIO with elotuzumab, pomalidomide and dexamethasone; pomalidomide, bortezomib and dexamethasone; or carfilzomib and dexamethasone.
  • Overall survival data were described as immature, so the trial’s survival benefit is not yet established.
  • Pfizer has not yet provided the detailed efficacy results that it plans to present and publish.
