Zai Lab reports 81.8% ORR and plans global Phase 3 trial for zoci in extensive‑stage small‑cell lung cancer

Zai Lab presented Phase 1b data for its DLL3‑targeting ADC zocilurtatug pelitecan (zoci) combined with atezolizumab in first‑line extensive‑stage small‑cell lung cancer. At the recommended Phase 3 dose of 1.6 mg/kg, the doublet achieved an objective response rate of 81.8% and a six‑month progression‑free survival rate of 81.9% among 33 patients. The regimen was well tolerated, with grade ≥3 adverse events in 9.1% of patients, and 66.7% of the 15 patients with at least six‑month follow‑up remained progression‑free. The company said it will launch a global randomized Phase 3 study by year‑end 2026.

The company says the data support advancing a platinum‑free regimen and the planned Phase 3 trial could unlock future revenue if efficacy is confirmed. Investors may view the strong early efficacy and safety as a catalyst for the stock.

  • 1Confirmed objective response rate was 81.8% (27/33) at 1.6 mg/kg zoci plus atezolizumab.
  • 2Six‑month progression‑free survival rate was 81.9% (n=33) for the same cohort.
  • 3Grade ≥3 adverse events related to zoci occurred in 9.1% (3/33) of patients.
  • 4Among 15 patients with ≥6‑month follow‑up, 10 (66.7%) remained free from disease progression or death.
  • 5A global, randomized Phase 3 study is expected to initiate by year‑end 2026.
  • 6In a triplet arm (zoci + atezolizumab + carboplatin, n=13) the objective response rate was 92.3% with higher grade ≥3 adverse events (76.9%).
