TG Therapeutics reports lower five‑year disability progression for BRIUMVI versus ocrelizumab and ofatumumab

TG Therapeutics released a five‑year matching‑adjusted indirect comparison of its anti‑CD20 therapy BRIUMVI against ocrelizumab and ofatumumab in relapsing multiple sclerosis. The analysis, using 545 BRIUMVI patients from the ULTIMATE I/II trials, found a 44% lower hazard of 24‑week confirmed disability progression versus ocrelizumab and a 49% lower hazard versus ofatumumab. Estimated rates of confirmed disability progression at 240 weeks were 9.3% for BRIUMVI compared with 16.1% for ocrelizumab and 9.4% for BRIUMVI versus 17.9% for ofatumumab. The study was funded by Neuraxpharm, TG’s European partner, and the results are described as exploratory pending direct head‑to‑head trials.

The company says the findings support BRIUMVI’s durable efficacy and could help it gain market share among anti‑CD20 options, which may influence future sales forecasts.

  • 1The analysis used individual data from 545 BRIUMVI‑treated participants in the Phase 3 ULTIMATE I and II trials.
  • 2BRIUMVI showed a 44% lower relative hazard of 24‑week confirmed disability progression versus ocrelizumab.
  • 3BRIUMVI showed a 49% lower relative hazard versus ofatumumab.
  • 4At 240 weeks, the estimated rate of confirmed disability progression was 9.3% for BRIUMVI and 16.1% for ocrelizumab.
  • 5At 240 weeks, the estimated rate was 9.4% for BRIUMVI and 17.9% for ofatumumab.
  • 6Annualized relapse rates for BRIUMVI declined from 0.118 in Year 1 to 0.032 in Year 5 versus ocrelizumab, and from 0.116 to 0.030 versus ofatumumab.
