Merck and Moderna report 42% lower melanoma recurrence risk with intismeran plus Keytruda in Phase 3 trial

Merck (MRK) and Moderna (MRNA) announced that their personalized mRNA cancer vaccine, intismeran, combined with Merck's Keytruda, cut the risk of recurrence or death by 42% in patients with completely resected stage IIB‑IV melanoma. The Phase 3 INTerpath‑001 trial also showed a 38% reduction in distant metastasis or death. The companies said they will now engage regulatory authorities about the data.

The company says the data position the intismeran‑Keytruda combination for adjuvant melanoma treatment, potentially expanding Keytruda's approved indications and creating a new revenue stream for both firms.

  • 1Intismeran plus Keytruda reduced the risk of recurrence or death by 42% (hazard ratio 0.58, 95% CI 0.45–0.75) versus Keytruda alone.
  • 2Intismeran plus Keytruda reduced the risk of distant metastasis or death by 38% (hazard ratio 0.62, 95% CI 0.45–0.85) versus Keytruda alone.
  • 3The primary analysis was based on an intention‑to‑treat population after a median follow‑up of 24.6 months.
  • 4Merck said it will engage with regulatory authorities based on the Phase 3 results.
  • 5Merck reported the same 42% reduction in recurrence/death risk and 38% reduction in distant metastasis/death risk.
